L'éthoxyrésorufine-o-déethylase, les adduits à l'ADN et les mocronuclei dans les organismes marins. Application à la surveillance des effets biologiques sur les côtes françaises ArchiMer
Burgeot, Thierry.
Three biomarkers [ethoxyresorufm-o-deethylase (EROD), DNA adducts and micronuclei (MN) were studied for biomonitoring applications along French coasts. EROD was developed at an experirnental stage in two pilot sites, Seine Bay and Fos sur Mer, using the dab (Limanda limanda), dragon et (Callionymus lyra), red mullet (Mullus barbatus) and comber (Serranus hepatus) as target species. Higbest EROD activity was observed in the subestuarial part of Seine Bay and at the stations closest to sources of pollution from Cortiou and Fos sur Mer. The suitable number of fish for detection of a significant difference between two EROD activities was deterrnined as a function of sampling variance and a preselected statistical power. A preliminary study on hepatocytes in a...

Colocalization of coilin and nucleolar proteins in Cajal body-like structures of micronucleated PtK2 cells BJMBR
Silva,N.P.; Christofolini,D.M.; Mortara,R.A.; Andrade,L.E.C..
Cajal bodies (CB) are ubiquitous nuclear structures involved in the biogenesis of small nuclear ribonucleoproteins and show narrow association with the nucleolus. To identify possible relationships between CB and the nucleolus, the localization of coilin, a marker of CB, and of a set of nucleolar proteins was investigated in cultured PtK2 cells undergoing micronucleation. Nocodazol-induced micronucleated cells were examined by double indirect immunofluorescence with antibodies against coilin, fibrillarin, NOR-90/hUBF, RNA polymerase I, PM/Scl, and To/Th. Cells were imaged on a BioRad 1024-UV confocal system attached to a Zeiss Axiovert 100 microscope. Since PtK2 cells possess only one nucleolus organizer region, micronucleated cells presented only one or...

Tomato and garlic by gavage modulate 7,12-dimethylbenz[a]anthracene-induced genotoxicity and oxidative stress in mice BJMBR
Bhuvaneswari,V.; Velmurugan,B.; Abraham,S.K.; Nagini,S..
Chemoprotection by dietary agents is a promising strategy for cancer prevention. The aim of the present study was to evaluate the combined effect of tomato and garlic against 7,12-dimethylbenz- [a]anthracene (DMBA)-induced genetic damage and oxidative stress in 12-14-week-old male Swiss albino mice. The animals were randomized into experimental and control groups and divided into eight groups of five animals each. Group 1 animals were injected intraperitoneally with 35 mg/kg body weight DMBA suspended in peanut oil as a single dose. Groups 2-4 animals received tomato (500 mg/kg body weight), garlic (125 mg/kg body weight) and a combination of tomato and garlic for 5 days by gavage, respectively, followed by DMBA 1.5 h after the final feeding. The doses of...

Chemoprotective effect of cysteamine against the induction of micronuclei by methyl methanesulfonate and cyclophosphamide Genet. Mol. Biol.
Santos-Mello,Renato; Deimling,Luiz Irineu; Lauer Júnior,Claudio; Carvalho,Thaís Rieger de.
Cysteamine or 2-mercaptoethylamine (MEA) is an aminothiol with a well-known radioprotective action. No specific information is available in the literature about the possible chemoprotective action of MEA against genotoxic chemical agents. This paper presents the results of studies on the ability of MEA to protect mouse bone marrow polychromatic erythrocytes against the induction of micronuclei by alkylating agents such as methyl methanesulfonate (MMS) and cyclophosphamide (CP). We observed that MEA administered intraperitoneally 30 min before or 30 min after the administration of MMS or CP significantly reduced the frequency of micronucleated polychromatic erythrocytes (MNPCEs) induced by the alkylating agents. When MEA was administered in combination with...

Micronuclei formation in liver fibrosis samples from patients infected by hepatitis C virus Genet. Mol. Biol.
Almeida,Terezinha M.B. de; Leitão,Regina Maria C.; Carrilho,Flair J.; Sonohara,Shigueko.
Genetic research on fibrosis outset and its progression in chronic hepatitis (CH) by hepatitis C virus (HCV) are limited. The lack of cytogenetic data led us to investigate the presence of micronuclei (MNi), as a sign of genomic damage. Hepatocytes of hepatic parenchyma from 62 cases diagnosed with CH associated with HCV and displaying different degrees of fibrosis (F1-F4) were analyzed. These data were compared to 15 cases without fibrosis (F0). Twelve healthy liver parenchyma samples were included as control. All samples were obtained from paraffin-embedded archival material. Micronucleated hepatocytes (MN-Heps) were analyzed through Feulgen/Fastgreen staining. Results showed that the rates of MN-Heps in the F4 group were statistically significant (p...

Genotoxic effects of the diesel water-soluble fraction on the seahorse Hippocampus reidi (Teleostei: Syngnathidae) during acute exposure Rev. Bras. Zool.
Santos,Celina Alcoforado; Novaes,Larissa Simões; Gomes,Levy Carvalho.
The high toxicity of diesel components makes oil spills a threat to the biota in coastal marine environments. The genotoxic effect of the diesel water-soluble fraction (DWSF) on Hippocampus reidi (Ginsburg, 1933) was assessed. Fish were exposed to three different DWSF dilutions for up to 96 hours, and genotoxicity was analyzed using the micronuclei test and the comet assay. The micronuclei test revealed no significant differences between any of the DWSF dilutions and the control in the 24-hours period; however, micronuclei increased in fish exposed to 1:500 and 1:100 DWSF dilutions for 96 hours. For all dilutions, there was an increase in micronuclei in fish exposed for 96 hours when compared to those exposed for 24 hours. The tested dilutions increased...

Cytological and cytogenic effects induced by thiabendazole on Allium cepa root meristem JBAG
Andrioli,Nancy B; Mudry,Marta D.
The toxicity and genotoxicity of thiabendazole (TBZ) in its commercial formulation Foldan® was studied in Allium cepa meristematic cells at concentrations ranging between 10 and 250 µg / ml. Our results show that exposure to TBZ induces a significant increase in the frequency of anaphase telophase chromosomal aberrations, micronuclei and binucleated cells at all the concentrations of TBZ assayed. The frequency of anaphase telophase aberrations was observed to reach a plateau at the concentration of 50 µg / ml of TBZ, which could be probably due to TBZ mitotic toxicity manifested at concentrations of 100 and 250 µg / ml. An increase was also observed in the rate of metaphase and anaphase, which was, in turn, indicative of alterations in chromatid...

Genotoxic effect of high nitrate water consumption in men and woman from northern Mar del plata, Argentina JBAG
Poli,M.N; López Miranda,L.A; Fernández Iriarte,P.J; Zanier,G.J; Iúdica,C.E.
Previous research has shown connections between the exposure to different agents and mutations in germinal and somatic cells. One of such agents, nitrate (NO3), can be potentially genotoxic for individuals who drink high nitrate well water, and may cause a subsequent impact on their health and on the whole ecosystem. The aim of this research was to conduct genotoxicity assays in somatic cells from people living in the northern area of Mar del Plata city, Argentina. The purpose of these experiments was, in turn, to establish the possible relationships between mutagenic effects and high nitrate water consumption. To this end, different diagnostic tests were carried out to detect potential genetic and/or chromosomal alterations. A non-exposed population equal...

Micronucleated lymphocytes in parents of Down syndrome children BJMBR
Silva-Grecco,R.L.; Navarro,G.C.; Cruz,R.M.; Balarin,M.A.S..
Down syndrome (DS) is the most common disease due to an autosomal aneuploidy in live born children and also the major known genetic cause of mental retardation. The risk of a DS pregnancy increases substantially with increasing maternal age. However, several women aged less than 35 years at conception have a child with DS. The micronucleus (MN) assay can identify chromosome breakage or chromosome malsegregation and is an ideal biomarker to investigate genomic instability. The aim of the present study was to determine the frequency of peripheral lymphocytes with MN in the parents of DS individuals. The subjects were 17 couples, 1 father and 9 mothers, and 24 couples who had at least one healthy child formed the control group. For each individual we...

Protective effects of steroidal alkaloids isolated from Solanum paniculatum L. against mitomycin cytotoxic and genotoxic actions Anais da ABC (AABC)
VIEIRA,PABLINE M.; MARINHO,LORENA P.M.; FERRI,SUZANA C.S.; CHEN-CHEN,LEE.
Solanum paniculatum L. is a plant species widespread throughout tropical America, especially in the Brazilian Cerrado region. It is used in Brazil for culinary purposes and in folk medicine to treat liver and gastric dysfunctions, as well as hangovers. Previous studies with S. paniculatum ethanolic leaf extract or ethanolic fruit extract demonstrated that they have no genotoxic activity neither in mice nor in bacterial strains, although their cytotoxicity and antigenotoxicity were demonstrated in higher doses. In order to assess the possible compounds responsible for the activities observed, we fractionated the ethanolic fruit extract of S. paniculatum, characterized by 1H and 13C NMR spectra, and evaluated two fractions containing steroidal alkaloids...

Effects of cadmium on root growth, cell division and micronuclei formation in root tip cells of Allium cepa var. agrogarum L Phyton
Wang,QL; Zhang,LT; Zou,JH; Liu,DH; Yue,JY.
The effects of various cadmium (Cd) concentrations (10, 50 and 100 μM) on root growth, cell division and micronuclei formation in root tip cells of Allium cepa var. agrogarum L. were investigated to better understand the processes of Cd-induced apoptosis. The results indicated that 10 μM of Cd had an obviously passive influence on root growth during the 24 h treatment, and that the influence was even more serious with increasing Cd concentrations and duration of treatments. The mitotic index decreased with increasing Cd concentration and duration of treatments. Cadmium induced c-mitosis, chromosome bridges, chromosome stickiness and micronuclei. The frequency of cells with these disorders varied with prolonging the duration of treatments...
